A diagram illustrating the differences in the DNA microarray process and RNA sequencing (RNA-Seq). DNA microarrays consist of nucleic acid probes affixed to a surface. RNA samples are converted into complimentary DNA (cDNA), and are then labeled with fluorescent tags (1). The labeled cDNA fragments hybridize the nucleic acids on the array (2), and levels of gene expression are measured by scanning the fluorescence levels of the microarray (3). In RNA-Seq, samples of RNA are fragmented and converted into double-stranded cDNA with additional sequencing adaptors (A). The cDNA library is sequenced (B), and the results are mapped to the genome (C).
